Reboot is always necessary and your networkcard must support it !

In 2000 : go to the settings of your networkcard and put in a new address in the advanced tap local administration adress or network address if mentioned.

In NT4 : regedit, in hkey_local_machine/system/currentcontrollset/services//parameters add new stringvalue NetworkAddress and as value, type your new wanted MaAddress
